Ethan Shaw
When Ethan was a kid growing up, he was always out in the sun. He worked outside and didn’t protect his skin from harmful UV radiation. Years later, he developed scaly patches on his head that later progressed to cutaneous squamous cell carcinoma, the second most common type of skin cancer. Surgeries, skin grafts, and chemotherapy failed to prevent his tumors from spreading. Concerned about his future and having exhausted standard treatment options, Ethan enrolled in a clinical trial testing a PD-1 blocking antibody called cemiplimab. The drug eliminated all the tumors on the outside of his head in about six weeks. Today, Ethan has no evidence of cancer and is able to enjoy his life with his wife, Donna, and their two children.

In 2004, at only 22 years old, Sharon Belvin was diagnosed with stage 4 melanoma. She was just completing graduate school in elementary education and preparing for her upcoming wedding when she received the life-altering news. For Bob Carlson, 71, the standard treatment for his stage 4 lung cancer was almost worse than the disease itself. The 16 months of chemotherapy made him so sick he almost didn’t want to live. A search for better options eventually led him to Dr. Roy Herbst at Yale Cancer Center, who was conducting a clinical trial of a new immunotherapy drug called MPDL3280A. This drug, made by the biotech company Genentech, belongs to a class of immunotherapy drugs called checkpoint inhibitors; by “taking the brakes off” the immune system, the drug enables a more powerful attack against cancer.

Just before Christmas in 2014, Debbie Guadagno was looking forward to spoiling her grandkids with presents and spending time at family events. She had no idea that a tumor the size of a football was growing on her kidney until a routine blood test showed elevated calcium levels and she was sent for further testing. Scans showed that the cancer had spread to her liver, lung, and spine. With the help of her husband, Gary, and sister, Cindy, Debbie made it to all her appointments, receiving treatment for two years. After more than four years after her diagnosis—she was declared cancer-free by her medical team.
